Analysis of Somatic Mutation in Redifferentiated Plants by Electrophoresis of Saintpaulia ionantha Wendl . Cultured in vitro

Abstract

|
The present studies concerned with some efforts to elucidate certain environmental factors involved in occurrence of somatic mutation in Saintpaulia cv. Delaware and cv. Julianne during the callus growth and organ differentiation when in vitro tissue cultured, and attempts were made as to suppress the mutability. Mutation-lessening Ca, myo-inositol and pectin were added to MS cultural media at varied concentration levels were assessed by means of electrophoretic analysis of the extent of mutated clones. Few observations as the result were noted as shown below;
Esterase and phosphatase zymograms for the culture tissue containing more Ca with myo-inositol revealed a very distinguishable difference in the band patterns when compared with mother plant. It was presumed that a reduction in occurrence of mutation is due largely to the Ca protective function at the primary cell wall of differentiating plant tissues and probably DNA structures during the course of in vitro culture.
